目的探討TLSFJM(JM急性T淋巴細胞白血病細胞分泌的抑制因子)作為抑制因子對小腸移植急性排斥反應的抑制效應及其機理,并與FK506的抑制效應特點和副作用進行比較分析.方法雄性BN和LEW大鼠分別作為供、受體行小腸移植,共分5組: 小腸移植組(SBT組)、大劑量FK506 [0.5 mg/(kg·d)]組、小劑量FK506 [0.25 mg/(kg·d)]組、TLSFJM [10 U/(kg·d)]組和FK506 [0.25 mg/(kg·d)]+TLSFJM [10 U/(kg·d)]組.FK506和TLSFJM分別以肌肉或腹腔注射方式給藥.在不同時間段分別觀察各組動物體重、生存時間及肝、腎功能,組織病理學檢查排斥反應發生情況.結果 TLSFJM應用7 d,對移植大鼠肝、腎功能無損害.TLSFJM+小劑量FK506不但避免了大劑量FK506對肝功能的損害,而且顯著延長了受體的生存時間.但單獨應用TLSFJM仍有一定程度排斥反應出現,不能明顯延長受體的生存時間.結論 TLSFJM作為一種有效的移植免疫抑制因子,聯合小劑量FK506應用能延長宿主和移植腸的生存時間,延緩排斥反應的發生,減輕排斥反應的強度,避免大劑量FK506治療帶來的并發癥.TLSFJM有望成為一種新型、高效、低毒的免疫抑制劑.
