目的:探討重組人IGF-1和生長抑素-14單獨及聯合應用對甲狀腺相關眼病患者眼眶成纖維細胞增殖的影響。方法:體外培養甲狀腺相關眼病患者眼眶成纖維細胞,采用四甲基偶氮唑鹽(MTT)比色法檢測不同濃度及作用時間的重組人IGF-1和生長抑素-14對細胞增殖的作用。結果:50μg/L以上濃度IGF-1對眼眶成纖維細胞有促增殖作用,5nmol/L以上濃度的SST對眼眶成纖維細胞的生長有抑制作用,均呈量效和時效關系。IGF-1和SST聯合應用時, 50nmol/L以上的SST能阻斷100μg/L的IGF-1促細胞增殖的作用。結論:IGF-1可刺激眼眶成纖維細胞增殖。SST能抑制眼眶成纖維細胞增殖,并能阻斷IGF-1促進細胞增殖的作用,此抑制作用與濃度有關,提示SST能直接通過受體后機制抑制IGF-1誘導的眼眶成纖維細胞增殖。
