        • Endocrinology and lipid metabolism

Endocrinology is closely related to lipid metabolism. Lipotoxicity affects the abnormal function of various endocrine organs, and leads to diabetes, fatty liver, metabolic syndrome and other endocrine and metabolic diseases. It is an important strategy to prevent the lipid toxicity. Endocrine disorders can also cause dyslipidemia. Studies have found that thyroid and gonadal glands play an important role in lipid metabolism. Their molecular mechanisms are gradually revealed and will be a new therapeutic target for dyslipidemia. Lipid metabolism disorders play an important role in the development of endocrine and metabolic diseases.

        • The protective effects and mechanism of erythropoietin on human retinal pigment epithelial cells during oxidative injury

Objective To investigate the protective effect and mechanism of erythropoietin (EPO) on injury of human retinal pigment epithelial (hRPE) cell induced by hydrogen peroxide (H2O2). Methods Take subcultured hFRPE cells as study target. They were treated with 800 mu;mol/L of H2O2 for 3 hours to establish the cell injury model. The cultured cells were divided into three groups:control group, simply injury group and therapeutic group which again divided into 10 IU/ml, 20 IU/ml, 40 IU/ml,60 IU/ml subgroups according to the concentration of recombinant human erythropoietin(rhEPO). NF-kappa;B was measured by immunohistochemistry. The content of Malondialdehyde(MDA) which was the product of cellular lipid peroxidation and the releasing rate of lactate dehydrogenase(LDH)were estimated by chromatometry. Results H2O2 could elevate the level of MDA and the releasing rate of LDH, compared simply injury group with control group, the differences were significant.(tLDH=29.746,tMDA=20.426,Plt;0.05); Compared all of therapeutics groups with simply injury group, the releasing rate of MAD and LDH were decreased obviously, the differences were significant.(LDH t10IU=5.770,t20IU=12.774,t40IU=19.818,t60IU=24.833,Plt;0.05;MDA t10IU=5.345,t20IU=10.278,t40IU=18.571,t60IU=20.247,Plt;0.05); The correlative analysis results of each therapeutic subgroup were: ①the concentration of rhEPO had negative correlation with the relation rate of LDH and the content of MDA(r=-0.976,P=0.024; r=-0.968,P=0.032) ; ②the concentration of rhEPO had positive correlation with the nuclear translative rate of NF-kappa;B(r=0.998,P=0.002); ③the nuclear translative rate of NF-kappa;B had negative correlation with the content of MDA(r=-0.954,P=0.046). Conclusion EPO can protect hFRPE cells from the injury of H2O2, the mechanism may be related to the activation of NF-kappa;B.

        • CHANGE OF SUPEROXIDE DISMUTASES AND LIPID PEROXIDATION IN RETINA WITH PHOTOCHEMICAL INJURY

Wistar rats weaned were raised through 10 weeks under cyclic illumination of 12 hours light and 12 hours darkness, with four different fluorescent colour lighting condition:75 lx and 300lx blue light,300 lux white and 300lux pink light to study the change of superoxide dismutases(SOD)and lipid peroxied(LPO)in the retina. This paper shows that photic oxidative reaction reduces SOD in the retina and oxidizes polyunsaturated fatty acids to become LPO and that complex visible light oxidizes retina easier than simple wave lengths visible light does. The shorter the wave lengths of visible light is and the brighter the illumination is the more serious the oxidative damage of the retina is. • EXPERIMENTAL STUDY ON THE CHANGE OF HEPATOCYTE MITOCHONDRIAL CALCIUM CONTENT AND LIVER LIPID-PEROXIDATION AFTER BILIARY OBSTRUCTION

To investigate the mechanisms of hepatic injury after biliary obstruction. After a rat model of complete biliary obstruction(CBO) was induced, hepatocyte mitochondria was isolated and the calcium content of mitochondria, the contents of liver malondialdyhyde (MDA) and superoxide dismutase (SOD), the levels of serum T-Bil, ALT, ALP and GGT were measured in each group. Results: After CBO, mitochondrial calcium content, liver MDA and serum T-Bil, ALT, ALP, GGT became increased progressively, compared with control group (P<0.05); the liver SOD was decreased markedly (P<0.05). Mitochondrial calcium content was highly positively correlated with liver MDA content, serum ALT and ALP, r values were 0.967, 0.924 and 0.919 respectively (P<0.01). The liver MDA content was highly positively correlated with serum ALT and ALP, r values were 0.949 and 0.843 respectively (P<0.01). Conclusions: Mitochondrial calcium overload and liver lipid peroxidation may be the important mechanisms of hepatic injury induced by biliary obstruction.

        • EXPERIMENTAL STUDY ON THE ROLE OF LIPID PEROXIDES DURING LUNG INJURY INDUCED BY ACUTE CHOLANGITIS OF SEVERE TYPE

The contents of lipid peroxides(LPO)and vitamin E(V.E)and some functional index and histologic changes in the lungs from the the rabbit models of acute cholangitis of severe type(ACST)were measured dynamically. The results revealed that the V.E content decreased strikingly from 6 hours and the LPO level increased progressivelg from 12 hours in the lungs.Simultanuosly, the congestion and neutrophil infiltreation in the lung mesenchyme, and the endothelial cell damage and thrombosis in the lung blood capillaries had been observed. These suggest that acute lung injury induced by ACST is referable to the lipid peroxidation damage to the lung blood capillaries which is due to increased LPO and decreased antioxidants including V.E.

        • Role of CYP2C9 polymorphism in valproate-related lipidmetabolic in epilepsy children

Objective To investigated the influence of the CYP2C9 polymorphism on lipid profile and blood concentration in epileptic children with VPA. Methods This study collected the information of healthy children and epilepsy children who were treated with VPA in the First Affiliated Hospital of Putian University during June, 2018 to March, 2021. The serum lipids of 184 cases were collected and compared between epilepsy group before and after treatment with VPA with the control group. The polymorphism of CYP2C9 gene in children with epilepsy was detected, and lipid and VPA concentration were compared after classification. Results There was no significant difference in lipid between the control group and the epilepsy group before treatment (P>0.05); The TC, HDL, LDL, TC/HDL, LDL/HDL were statistically different in VPA treatment group from the control group (P<0.05), and there were statistical differences in TG, LDL, TC/HDL, LDL/HDL between the trial group before the initiation and VPA treatment (P<0.05); There is no correlation between VPA blood concentration and lipid (P>0.05). VPA concentration, TC, HDL, LDL, TC/HDL and LDL/HDL in CYP2C9 wild-type were statistically different from heterozygous mutant. Conculsions CYP2C9 polymorphism and long-term use of VPA caused the changes in serum lipid levels in epilepsy children.

        • Research on the Correlation between Lipid Accumulation Product and the Risk of Ischemic Cardiovascular Disease in Middle-aged and Elderly People of Chengdu Area

ObjectiveTo investigate the correlation between lipid accumulation product (LAP) and risk of ischemic cardiovascular disease (ICVD). MethodsThis cross-sectional study was performed among community residents from an urban community in Chengdu area between September 2011 and June 2012. Questionnaire survey was carried out. Each individual underwent biochemistry analysis and physical examination. In addition, brachial-ankle pulse wave velocity (BaPWV) and augmentation index (AI) were detected. Pearson correlation analysis was performed to explore the relationship between LAP and each cardiovascular risk factor. Liner regression model was used to analyze the relationship between LAP and ICVD. ResultsA total of 780 individuals with complete data were included in the analysis. LAP was correlated with blood pressure, total cholesterol, high density lipoprotein cholesterol, fasting blood glucose, and BaPWV (P<0.05). LAP was associated with the risk of ICVD (r=0.253, P<0.001). After being adjusted with sex, age and other cardiovascular risk factors, LAP was also correlated with the risk of ICVD (r=0.050, P<0.001). ConclusionsHigh LAP is associated with elevated cardiovascular risks and subclinical vascular damage. In addition, LAP is correlated with ICVD risk, thus it may be used to predict the incidence of ICVD to some extent. However, as the correlation is weak, our study does not support the direct use of this indicator to predict ICVD. Large-sample studies based on different races and ages are needed.

        • APOLIPOPROTEIN E GENE POLYMORPHISMS,DYSLIPIDEMIA AND CHOLECYSTOLITHIASIS

The present study was designed to elucidate the role of apoE polymorphism in the lithogenesis of cholecystolithiasis and to explore the hereditary pathogenesis of the disease. Polymerase Chain Reaction (PRC) was used as researching apoE phenotypes and allele frequencies in patients with gallstones (n=87) and in controls (n=50), and the fasting serum lipids of subjects were also measured. The characteristics of lipid variants were analysed among the patients with different apoE phenotypes. The results showed that the levels of TG (1.43mmol/L), VLDL-C(0.68mmol/L) in E2/3 patients were greatly higher than those in E2/3 controls (1.06mmol/L, P<0.05 and 0.48mmol/L, P<0.05), and LDL-C (1.41mmol/L) was markably lower in E2/3 patients than that in controls (2.04mmol/L, P<0.05). The levels of serum lipids decreased significantly in E3/3 patients with HDL-C (0.89mmol/L), HDL2-C (0.49mmol/L), HDL3-C (0.39mmol/L), and compared with those in E3/3 controls (1.28mmol/L P<0.05, 0.73mmol/L P<0.001 and 0.55mmol/L P<0.001). In E3/4 patients there were only slight changes of VLDL-C, LDL-C level. The results suggest that the average level of serum lipids in the same apoE phenotype patients with gallstones is higher than that in controls, and the different apoE phenotypes patients with gallstones have different characteristics of dyslipidemia. ε2 allele is probably one of the dangerous factor in the lithogenesis of cholecystolithiasis.
