        • STUDY ON THE EXPRESSION OF p16 AND Rb IN THYROID CARCINOMA

          【Abstract】Objective To demonstrate the role of p16 protein and Rb protein in the formation and progression and their relationship in thyroid carcinoma. Methods p16 and Rb from 46 patients with thyroid cancer were detected by SP immunohistochemistry. Results In thyroid carcinoma,the p16 positive rate was decreased with the decline of differentiation. There was an inverse correlation between the rates of p16 positive expression and involvement of lymph node,clinical stage and prognosis index.The Rb protein was not present in any cases as a nucleus staining. Conclusion The p16 gene is likely to play an important role in the genesis and malignant progression of thyroid carcinoma,while the Rb gene is not .

        • Role of Methylation of p16 Gene in Breast Cancer

          目的研究p16基因甲基化在乳腺癌中的分布情況,并探討其與乳腺癌發生、發展的關系。方法采用甲基化特異的PCR技術檢測四川省腫瘤醫院乳腺科2008年3~9月期間收治的38例原發性乳腺癌組織及距離腫瘤>5 cm以遠的正常乳腺組織中p16基因啟動子區CpG島甲基化頻率。結果乳腺癌組織中p16基因啟動子區CpG島甲基化頻率〔31.6%(12/38)〕明顯高于其正常乳腺組織〔5.3%(2/38)〕,差異有統計學意義(P=0.003)。有淋巴結轉移的乳腺癌患者中p16基因甲基化頻率〔45.5%(10/22)〕高于無淋巴結轉移患者〔12.5%(2/16)〕,差異有統計學意義(P=0.044)。結論p16基因啟動子區CpG島甲基化可能在乳腺癌發生、發展過程中起著重要作用。

        • 外源性p16基因對食管癌患者癌細胞系的生長抑制作用

          目的 觀察外源性p16基因質粒導入食管癌患者食管鱗癌細胞系Ec109后的表達及其對Ec109細胞增殖生長狀況的影響。方法 根據轉染質粒的不同或是否行p16 cDNA質粒轉染分為三組,每組分別為5個樣本。PcDNA3-P16轉染組(P16轉染組):采用脂質體(Lipofectamine)為載體,將PcDNA3-P16表達質粒導入Ec109細胞;空載體轉染組:用PcDNA3-neo空載體以上述相同方法轉染,作空白對照;未轉染組:未用PcDNA3-P16質粒轉染,作陰性對照。應用Northern斑點雜交、免疫組織化學、免疫熒光檢測轉化細胞的P16蛋白變化;流式細胞儀分析細胞周期變化,DNA片段化電泳檢測細胞凋亡。結果 P16轉染組外源性p16基因在Ec109細胞中表達后,細胞生長速度減慢,克隆形成能力下降,瞬時表達時有細胞凋亡發生,穩定表達后細胞存在G1期阻滯;空載體轉染組和未轉染組均未觀察到以上結果。結論 脂質體介導的外源性p16基因轉染對Ec109細胞有生長抑制作用,p16基因瞬時表達可誘導細胞凋亡。

        • Deletion and Mutation of p16/mts1 Gene in Human Gastric Cancer of Different Periods

          ObjectiveTo determine the frequency and type of p16 gene homozygous deletion and mutation with the progression of human gastric carcinogenesis.MethodsPCR (polymerase chain reaction),SSCP (single strand conformation polymorphism) and DNA sequencing were performed on 50 operative tissues, which included 7 early gastric cancer and 43 advanced gastric cancer. In this group there were 36 male and 14 female patients aged 35 to 68,which included 30 welldifferetiated adenocarcinoma and 20 poordifferentiated carcinoma; 19 patients with lymph node metastases.ResultsTotal mutation frequency of p16 gene was 8.00%(4/50), including 14.29%(1/7) of early cancer and 7.00%(3/43) of advanced cancer,there was no significant difference between early and advanced cancer (P>0.05). In addition, p16 gene mutation was not related to site,location,grade of tumor differentiation and lymph node metastases, respectively. Total deletion frequency of p16 gene was 16.00%(8/50), of which 18.60%(8/43) was detected in advanced cancer, but no cases in early cancers. There was a significant difference between early and advanced cancer (P<0.05), and frequency p16 gene homozygous deletion was related to grade of tumor differentiation and lymph node metastases, respectively. Direct DNA sequencing displayed that 4 point mutations detected in 99,125,433,441 base, there were 3 missense mutation and 1 samesense mutation, no deletion or insertion was found. Conclusionp16 gene mutation is an early event in gastric carcinogenesis,it helps early diagnosis of human gastric carcinoma. p16 gene homozygous deletion is a late event in gastric carcinogenesis,it might relate to metastases and recurrence of gastric cancer.

        • 食管癌p16基因變異分析

          目的 為了探討p16基因與食管癌發生、發展的關系.方法 應用聚合酶鏈反應、單鏈構象多態分析(PCR-SSCP)方法檢測30例食管癌中p16基因的純合性缺失和突變情況,并分析p16基因變異與食管癌的病理關系.結果 30例食管癌患者中4例為p16基因純合性缺失,2例存在突變.6例陽性樣品與其它樣品在年齡、飲酒史方面差異有顯著性(P<0.01),與淋巴結轉移、病理分期有差異(P<0.05);而與性別、腫瘤長度、腫瘤部位、病理類型無差異(P>0.05).結論 抑癌基因p16在食管癌中存在基因變異,其在食管癌的發生、發展中起重要作用.
